Bi-specific antibodies (BsAb) recognize and bind two different antigens or epitopes simultaneously and block two different signaling pathways to exert their effects. Compared with monoclonal Ab (mAb), which recognizes a single antigen, BsAb has many advantages. First, BsAb can redirect specific immune effector cells to adjacent tumor cells to enhance tumor killing, and BsAb can also increase binding specificity through the interaction of two different cell surface antigens. In addition, BsAb is able to simultaneously block two different pathways that perform distinct or overlapping functions in pathogenesis compared to drugs with single antibodies in combination therapies.
